N2 - Outcomes remain poor in half of melanoma patients despite standard checkpoint inhibitor treatments. The prevalence of melanoma-associated chondroitin sulfate proteoglycan 4 (CSPG4) antigen expression is about 70%, so effective immunotherapies against CSPG4 could benefit many patients. Because IgE has potent immunostimulatory functions in tissues, we are developing an IgE monoclonal antibody with human constant regions that recognizes CSPG4 to target melanoma. CSPG4-IgE binds to human melanomas, including metastases, mediates tumoricidal antibody-dependent cytotoxicity, and stimulates human IgE Fc receptor-expressing monocytes toward proinflammatory phenotypes. IgE exhibits antitumor activity in human melanoma xenograft models transplanted with human effector cells and is associated with increased macrophage infiltration, enriched monocyte and macrophage gene signatures, and proinflammatory signaling pathways in the tumor microenvironment. IgE prolongs the survival of patient-derived mouse xenografts reconstituted with autologous immune cells.
